The villagers probably wanted their next defender of their village to be a pure human, and not risk it turning on them as a cross breed. They also might think that having him and his sister have a child would make the child even stronger than Misaki, or as strong since they both have the same blood of past legendary warriors according to the wolf priestess in the beginning. So as far as I'm concerned they're probably not going to rape the sister, or at the very least their not going to impregnate her. This is probably for the best.
